在人类生存的环境中存在超过100,000种不同类型的真菌,其中双相性致病真菌有:皮炎芽生菌、荚膜组织胞浆菌、粗球孢子菌、巴西副球孢子菌、马尔尼非青霉菌和申克孢子丝菌。双相真菌的孢子入侵宿主体内后其形态便会由体外环境中非致病的菌丝形态转换为体内环境中致病的酵母形态。其中孢子丝菌病的发病率最高,可引起皮肤、皮下组织以及附近淋巴管的慢性感染,导致化脓、溃烂及渗出,偶可累及肌肉、骨骼、中枢神经系统等脏器。孢子丝菌病的病原体为申克孢子丝菌,具有双相性：25℃为菌丝相,不具致病性；37℃为酵母相,具有致病性。孢子丝菌由菌丝相转化为酵母相的过程被认为是孢子丝菌致病性形成的过程,对孢子丝菌形态转换过程的深入研究有助于揭示孢子丝菌病的发病机制。双相真菌的形态转化是一个多因素共同参与的复杂过程,是真菌通过其信号传导系统感受并将变化了的环境信号因子级联传递至细胞内,引起特定的基因表达进而行使功能的过程。既往的研究中本课题组曾应用比较蛋白质组学的研究方法比较了孢子丝菌菌丝相及酵母相的蛋白表达图谱,鉴定了24个酵母相特异或上调表达的蛋白质,其中包括双组份信号系统组氨酸激酶DRK1基因,为了进一步探讨DRK1在孢子丝菌双相转化和致病性形成中的作用,我们对此进行了研究。
